An event in your life that changed you Essay A new house, new school, and a completely new atmosphere were ahead of my nine-year-old life. I had lived in the same house for what it seemed like forever and had attended the same school since first grade. I knew everyone. From January to December, my calendars were filled with birthday parties I had to attend. Performing in talent shows with my best friends Dezerey, Jasmine, and Nykchasia were a yearly activity at Garden Valley Elementary School. The Robinson Centers summer camp was mandatory for my big sister, Breonna, and me. I was used to familiarities like the Chihuahua that ran to the front of its gate and barked every time I rode past it on my sleek, black rollerblades. Those rollerblades made me feel untouchable, at ease and confident until I had to ride them in a new area, surrounded by new, unfamiliar people. Close to the end of my fourth grade year, my mother told my sister and me that we were moving to Terricina Gold Apartments in Natomas. I did not think it was going to change anything. I was probably a bit excited for what was to come. I can clearly remember my first day at Two Rivers Elementary school being terrified. I believe that was when it first hit me that I was not at home anymore. My comfort zone left was tarnished. I remember having butterflies and feeling as if I was going to faint before I stepped foot onto school grounds. When I got to the blacktop all I could do was stand there in silence. The other kids were standing around in there cliques and it was obvious that the fifth graders controlled the far left gate on the black top. I remember one girl that stood out. She looked larger than life in my eyes. She was bright skinned, tall and everyone seemed to flock to her. She made me feel small without even knowing her. I was having trouble finding my classroom number on the blacktop where my class was supposed toà line up. I completely gave up after a few minutes mainly because I felt so out of place I just wanted to hide. Not to mention I am legally blind and even with my glasses I still cannot see all that well so finding my class line was a far reach for my nine-year old mind. I felt out of place and inferior so I I took it upon myself to run away and hide in the nearest bathroom. I stayed in that bathroom for a while, even after the bell rang. I eventually decided to take a step out of the bathroom and walk to my class. In fourth grade I absolutely did not have any social skills. Many children lived in my new apartments. I envied their large groups of friends and seeing them made me miss my old ones. I could never gather up the courage to talk to anyone. Thankfully, I had my older sister. She is the most outgoing between us and she managed to know everyone in nearly a week of our residency. I eventually met everyone that lived in the apartments but I never felt at ease with them as I did with my old friends. I rode my rollerblades all around those apartments and they made me happy like a piece of home was with me. I never realized that I was in fact an introvert. I suppose I had always been a timid person my mother brings up her memories of my behavior as a child often. I never realized my childhood behavior until now. I remember my mom changing my teachers and being too afraid to walk into the classroom even though I knew the teacher and all the students well. Moving away from my familiarities showed how quiet and unsociable I was. I did not have friends at school until I was well into my seventh grade year. Today, I am on the verge of twenty-years-of-age, and I still have to work on speaking up and being more sociable. Before I had many friends and after I moved, I did not. I believe if I stayed where I lived before I would have been given a sense of security. Looking back, I believe I would not have gained a great number of wonderful friends. My experiences with people in my apartments are always the topic of conversation with my family and friends and they bring on tons of laughs and feelings of embarrassment. I love revisiting my old Natomas neighborhoods and seeing my old friends. We always joke saying TG (Terricina Gold) for life. I guess you can call it a family.

However the large intestine is wider (about 3 inches) yet shorter than the small intestineà (in humans about 4. 9 feet in length as compared with 22 25 feet for the small intestine) and has a smooth inner wall. The longitudinal muscle of the muscularisà externa is concentrated into 3 bands called teniae coli. These 3 bands of muscle form pouches (haustra) because they are shorter than the rest of the colon. The epithelium of the mucosa is mostly goblet cells, and has a numerousà amount of crypts, there are no villi. (Martini, Ph. D. ) The ileocecal valve (sphincter) prevents materials from moving back into the ileum. In the upper half of the large intestine, enzymes from the small intestine complete the digestive process and bacteria produces the B vitamins (B12, thiamin, and riboflavin) as well as vitamin K. Martini, PhD) The large intestines primary function is to absorb water and electrolytes from digestive residues and storage of fecal matter until it can be expelled. The large intestine is the last attraction in digestive tube and the location of the terminal phases of digestion. In comparison to other regions of the tube, there are huge differences among species in the relative size and complexity of the large intestine. The most frequent and common disease of the large intestine is Crohns disease. The cause of Crohns disease is unknown. ( Britannica) Apart from the greater tendency for fistulas to form and for the wall of the intestine to thicken until the channel is obstructed, it is distinguishable from ulcerative colitis by microscopic findings. In Crohns disease, the maximum damage occurs beneath the mucosa, and lymphoid conglomerations, known as granulomata, are formed in the submucosa. Crohns disease attacks the perianal tissues more often than does ulcerative colitis. Although these two diseases are not common, they are disabling. ( Britannica) Because there is no specific etiology, a combination of anti-inflammatory drugs, including corticosteroids and aminosalicylic acid compounds, is used to treat Crohns disease. (Web MD) The drugs are effective both in treating acute episodes and in suppressing the disease over the long term. Depending on the circumstances, hematinics, vitamins, high-protein diets, and blood transfusions are also used. Surgical resection of the portion of the large bowel affected is often done. The entire colon may have to be removed and the small intestine brought out to the skin as an ileostomy an opening to serve as a substitute for the anus. In ulcerative colitis, as opposed to Crohns disease, the rectal muscle may be preserved and the ileum brought through it and joined to the anus. (Web MD) Lactose intolerance is the inability to digest significant amounts of lactose, the major sugar found in milk. Lactose intolerance is caused by a shortage of the enzyme lactase, which is produced by the cells that line the small intestine. Lactase breaks down milk sugar into two simpler forms of sugar called glucose and galactose, which are then absorbed into the bloodstream. Lactose intolerance is a problem caused by the digestive system. (lactose) People who do not have enough lactase to digest the amount of lactose they consume may feel very uncomfortable when they digest milk products.

Anti-tumour Immunity through GP-100-TLR Agonist Conjugation Enhancing anti-tumour immunity through gp-100-TLR agonist conjugation. Introduction Soluble cancer vaccines remain an area of high interest to researchers with the ability to enhance immune responses against present cancers and induce protective immunity against future cases. In developing new vaccines finding ways to increase the immunogenicity of cancer antigens is a major challenge(1-3). The addition of Toll-like receptor (TLR) agonists is one strategy which can successfully boost immune cell activation and response to cancer antigens. By stimulating TLRs, these agonists increase expression of several co-stimulatory molecules on antigen presenting cells (APCs) such as CD80/86 and CD40(4-6).They also increase tumour peptide loading onto type 1 2 Major histocompatibility complex (MHC) proteins. Together this leads to greater activation of tumour-specific effector immune cells such as CD4+ and CD8+ T-cells resulting in increased tumour clearance via their cytotoxic activity. Vaccine formulations which have included antigen and TLR agonists as a mixture have had pro mising results with many in clinical trials(4, 7, 8). Despite this, few have assessed the effect of chemically conjugating these constituents, a strategy which could increase efficiency of both TLR activation and peptide loading onto MHC(9-12). Many conjugation strategies that do exist today capitalise on the use of pH and redox sensitive linkers. Differences in pH and redox environments intracellularly enable triggerable release of these vaccines whilst protecting antigen and agonist from degradation extracellularly where they are administered. Research into the use of Glutathione-sensitive disulphide linkers has demonstrated that the immune response to model antigen Ovalbumin'(OVA) could be increased through linkage to the TLR agonist, CpG oligodeoxynucleotide (ODN)(10, 11). Our research aims to repeat this using both stable and reversible linkers as well as a more clinically relevant, tumour associated antigen (TAA) called gp-100 expressed on melanomas. In addition, we aim to ass ess the effectivity of different TLR agonists within conjugates including Polyinosinic polycytidylic acid (Poly I:C) and two different classes of CpG ODNs, B and C respectively. Each of these agonists activate different signalling pathways within antigen presenting cells leading to unique cytokine profiles and T-cell responses. Poly I:C for example, is a potent activator of TLR3 which activates the TRIF pathway inducing release of type 1 interferons such as IFN Beta(6, 13, 14). This increases MHC-I expression and stimulates a Th1 type immune response which favours cell-mediated immunity including CD8+ T-cell activation. In comparison, CpG class B and C stimulate TLR 9 activating the MYD88 pathway and release of proinflammatory cytokines IL-6 and IL-17. This results in enhanced CD4+ and CD8+ T-cell responses, B cell activation and antibody production(10, 11, 15). Both types of response have potential to give clinical benefit in different ways highlighting the potential of these conju gates in tumour treatment. Finally, we will also assess how the composition of the TAA effects its presentation on MHC. To assess this, a smaller Gp-100 peptide which does not require intracellular processing will be compared to a longer peptide requiring processing. This project will assess which conjugates enhance anti-tumour responses in mice and how they achieve this looking specifically at Dendritic cell activation and CD8+ T-cell proliferation and cytokine production. Hypotheses In terms of CD8+ responses, T cell proliferation and cytokine release, I hypothesize that Poly I:C reversibly linked to processed gp-100 will be the most effective inducing a strong Th1 response and IFN-B cytokine release. This is because Poly I:C stimulates several intracellular signals in addition to TLR3 including RIG-1 and MDA-5. This would lead to increased release of proinflammatory cytokines including IL-6, IL-12, IL-1B and IFN-B. Specifically the release of IFN-B would induce a strong anti-viral-like Th1 T cell response through increased expression of MHC-1 molecules on APCs and inducing release of IFN-Y, TNF-A and IL-2 from CD4+ T cells. As well as this factor, TLR3 is exclusively expressed on myeloid dendritic cells, the most effective dendritic cell subset in presenting antigen, and not expressed on plasmacytoid dendritic cells. Secondly, I Hypothesize that CpG class C will induce the most cytokine production in dendritic cells including the cytokines IL-6, IL-12, IFN-A and IL-1B. CpG molecules stimulate TLR9 which is expressed in the endolysosomal compartment of plasmacytoid dendritic cells exclusively. As a dendritic cell subset, plasmacytoid DCs are well known for their proinflammatory cytokine production at levels much higher than other DC subsets. CpG class C in particular stimulates the release of IFN-a in addition to IL-6, IL-12 etc. stimulating both a Th1 response and a B cell response. Aims and objectives Aims Produce gp-100-CpG ODN and gp-100-Poly I:C reversible and stable conjugates with either processed or non-processed Gp-100 peptides Measure dendritic cell subset activation through expression of MHC-II, CD40 and CD86 molecules and cytokine release (IL-12, IL-6, IL-1B, IFN-A and IFN-B ) Measure Tumour specific T cell activation (CD4+ and CD8+), proliferation using carboxyfluorecein succinimidyl ester (CSFE) and cytokine release (IFN-Y, IL-2,TNF-A) Methods The proposed project for the year will focus on three main objectives 1) Produce gp-100-CpG ODN and gp-100-Poly I:C reversible and stable conjugates with either processed or non-processed Gp-100 peptides. First, we will modify free amino groups on the lysine residues of each gp-100 peptide (processed amino acid sequence: KVPRNQDWL vs unprocessed: CAVGALKVPRNQDWLGVPRQL) and TLR agonists (suspended in a modification buffer ph. 8). Then we will link these together with either the stable linker (HYN) or the reversible linker (HYN-SS) in a ph. 6 conjugation buffer separately. Product concentration after each individual modification step will be measured using Nanodrop1000 at 280 m after desalting excess product using vivspin 500 filter. Final product conjugation will be confirmed using the reversed phase liquid chromatograph at the School of Pharmacy which will allow us to visualise each individual product according to their differing polarities, and quantify their ratio. Our second objective is to Measure dendritic cell subset activation through expression of MHC-II, CD40 and CD86 molecules and cytokine release (IL-12, IL-6, IL-1B, IFN-B, IFN-A). To achieve this, we will isolate bone marrow cells from C57BL/6 mice and treat with GM-CSF to produce CD11c+ dendritic cells. These will then be treated with either individual TLR agonists, TLR agonist-gp100 mixtures or TLR agonist-gp100 conjugates (reversible or non-reversible). After 24hrs of treatment these cells with be stained with fluorescent antibodies for CD80, CD40, CD11C, and MHC-II, viewed on the Gallios flow cytometer in Pathology and analysed using Kaluza software. This experiment will be repeated at least three times to enable statistical analysis, which will be performed using Graph Pad prism software. Cytokine release from these cells will be measured using an enzyme linked immunosorbent assay (ELISA) for IL-6, IL-12, IFN-B and IFN-a. Our third objective is to Measure Tumour specific CD3+ T cell: activation (CD8+), proliferation (CSFE) and cytokine release (IFN-Y, IL-2). This will be achieved through isolation of splenocytes from Pmel (T-cells specific to gp-100) transgenic mice and sorting of CD8+ cells using the Automacs machine at Pathology. These cells will then be stained using CSFE and co-cultured separately with C57BL/6 BMDCs treated according to objective 2. After 72hrs cells will be analysed using the Gallios flow cytometer to measure T-cell activation (CD3+) and proliferation (CSFE). To measure cytokine release, cell cultures will undergo an ELISA for IFN-Y and IL-2. Emerson Essay -- essays research papers fc
In Emerson's Self-Reliance we see the crowning work of the transcendentalist movement. In this piece Emerson explains his belief in the innate divinity of man and defines our "Self-Reliance" as the broad identity in which we personally participate. Emerson challenges his readers to not conform to traditional practices in a variety of realms. However, he punctuates just four aspects of these challenges to tradition and they are: religion, education, art, and society. I found these passages to be the best representatives of Emerson's ideology due to their poignancy and numbered paragraphs. He talks of these challenges to man as revolutions due to a greater self-reliance. The profoundness of thought in this piece is surprising to me given the historical period Emerson was bathed in. His thoughts on genius are the means of conveyance for his ideal about nonconformity and originality. Also, the personal and emotional connection with which Emerson uses to convey original thoughts and ideas is apparent in his evaluation of great minds of the past. The first passage regards the challenge to revolutionize religion; and more importantly, to discount the practice of prayer and creeds. He says, "Prayer that craves a particular commodity, anything less than all good, is vicious" (Robinson 102). He is basically announcing his contempt for the pious nature man has come to have and his belief that we should not pray for things we can achieve ourselves. He goes on to say, "But prayer as a means to effect a private end is meanness and theft. It supposes dualism and not unity in nature and consciousness" (Robinson 102). He is equating prayer with begging to God and believes it is not needed when you become one with God and therefore can see prayer in all productive actions. Prayer for Emerson creates a distinction between himself and God and does not allow for the self to become one with nature and consciousness. He also critiques man's practice of creeds and he believes the practice of one negates all others and vice versa. He says creeds are "a disease of the intellect" (Robinson 103). Whereas prayer is a disease of the will, creeds perform a habitual complacency in the life of man which allows only for the teachings of one particular idea. This is intellectual death for Emerson. To not only have to beli... ...hat is known is not valuable and not beneficial, and what is unknown is original, daring, valuable and great. The greatness resides inside of us and we must excavate it through constant reevaluation of our principles and virtues, without regarding foreign influences. In conclusion, I believe Emersonââ¬â¢s applicable challenges can be identified as his leading arguments when concerned with individual and personal revolution. His views on religion, education, art, and society are explicated through his gifted intuitional understanding and reason. By reasoning to the reader through vivid examples which are apparent and self-evident, he creates the proof for his understanding of reasonââ¬â¢s uses to question what we are perceived to know. The personal connection to Emerson is clear in his engaging emotional remark in which we can simply recognize as his affection for the original, misunderstood, and individual contributions great minds of the past have made. Causes of the French Revolution The French Revolution was essentially a class war between the emerging Bourgeoisie against the Privileged class, this meant they saw the Privileged class as the only hurdle between themselves and equality within French society. Many of the ideas they pursued stemmed from the enlightenment and they believed that in order to gain their full economic, social and political potential and gain equality, the Bourgeoisie had to eradicate the privileges that were halting their rise in society. To do this they had to seize power for themselves and gaining power within the government and making badly needed changes, such as, improving the tax system, creating a fair system of production where profits went to the producer, improving the whole fiscal system of the government, improving the geographical divisions of France and the problems they caused, plus many more. The revolution was a fight for equality and recognition by the Bourgeoisie, it was not a revolt against poverty, for many of the French people had been living in poverty for centuries and had learnt to live with it . France had prospered in the Eighteenth century. France had had no major famines or plagues, its population had increased, there had been no wars on its soil, industry such as textiles was doing well and offshore trading had increased enormously. It was the Bourgeoisie that had enabled this abundance of wealth and they were emerging as the economic power inside France. But their new found power was being smothered by the privileged class. The privileged class of nobility and clergy, who owned the bulk of the land, were using their ancient rights to plunder most of the profits that were made by the lower classes. This meant that the; Ã ¬Prosperity came only to those who held fairly large estates, who exercised feudal rights, or who could manipulate farm rents.Ã ®1 The only people who fell under these categories were the Nobles of the robe and the clergy. So in order to get the prosperity equally divided in society and thus profit themselves, the bourgeoisie had to get rid of the privileged classes ancient rights. To do this they had to revolt against the government, that supported the privileged classes and which was reluctant to make the necessary reforms. It is seen here that it was the Bourgeoisie were struggling against the Privileged class ... ...hed to them, because their privileges were taking large percentages of profits, adding on-costs to goods, causing massive inflation and reducing the wages of the middle class. rEsource and Learning Teams Essay -- Personal Perspective Work Resourc
rEsource and Learning Teams We are at the beginning of a journey of understanding. Understanding ourselves, other people, and the businesses we work for. We have many tools at our disposal for our journey. Two of the most important tools are rEsource and learning teams. In this paper I will discuss the values of these tools as well as the value of problem-based learning as it relates to our work and social lives. From the time I first logged on, I saw that rEsource would be our most valuable tool. The Value of rEsource The rEsource course page has many useful links to utilize throughout our journey. The e-Book and library links are excellent, with them research possibilities are endless. The link for the center of writing excellence is also a considerable commodity. With this link a writing tutor can be utilized as well as a writing style critic. With these sources I plan on honing my writing skills so that I not only have the business knowledge but I will also be able to present this knowledge in an organized and professional format. The rEsource page is very well organized and provides quick links to the weekly topics, readings, assignments, and quizzes. This comes in very handy for checking current status and staying on schedule. While these links are very valuable my peers are also a very valuable tool. Measurements of physical characteristics are used widely in everyday human life, whether in household or industrial processes, in service or in scientific researches. Some of the main categories of regularly measured parameters are weight, length, area, volume, temperature, density, concentration, voltage, amperage and power. Measurement units historically differed from nation to nation, but over time there is a straight tendency for unification of units. Metric system is recognized as universal in ever-growing number of countries and this allows to minimize differences and errors in international scientific, trade and other types of communications. In metric system, meter, its multiples and fractionals are used for the measurements of length, square meter and its derivates are for measurements of area, cubic meter ââ¬â for measurements of volume. For measuring temperature there are Celsius or Fahrenheit degrees used, for measuring density ââ¬â kilogram per cubic meter. Voltage in international metric system is measured in volts, amperage ââ¬â in amperes, and watts are used for measuring power. To perform measurements one needs to have appropriate instruments. These instruments should have accuracy grade acceptable for the type and purpose of measurements that will be performed. When measurement of length is performed, the only kind of instruments needed is tape-measure or straight scale, if size of measured object is relatively small. If distance that has to be measured is large, and surface between starting and finishing points is not smooth, more sophisticated instruments will be needed to perform required measurements. Measurements of area and volume will sometimes require certain calculations in addition to direct measurements. If measurement of an area of regular shape is performed, it is enough to measure length of the sides of that area and then apply relatively simple formulas for calculation of that area. But if the area measured has irregular shape with edges of bizarre form, calculations will be far more complex and may require solution of integral equations to determine approximate size of an area in question. When it is necessary to measure volume of a liquid, volumetric glassware is enough. But when the volume of a body with complex form shall be calculated, procedure of measurement may become much more complicated. If the body with unknown volume is available for direct measurement, it may be submerged into the water or any other liquid and volume of displaced liquid that is equal to the volume of that body, could be measured. But if that body is not available for direct measurement and is given in form of a scheme with known parameters measured or set beforehand, then rather complicated calculations may be required to estimate the volume of a said body. Density of a liquid may be measured using special instrument called densitometer, which is based on displacement of a certain amount of liquid by the weight of the instrument. The higher density of examined liquid, the smaller will be volume of liquid displaced. For estimation of the density of gases or solid bodies another methods shall be applied, because displacement-based densitometry cannot be applied to non-liquid substances. Measurements of mass and volume are usually performed, and density then is calculated using the simple formula of mass divided by volume. Temperature can be measured using thermometers of different types ââ¬â liquid-filled, electric, etc. Electric parameters like amperage, voltage and power, could be measured either directly using specific measuring devices, or indirectly by measuring related parameters and later calculating values of parameters required. For a list of physical parameters there are specific units in the system of measurements. Some of the examples were given above. For another example, unit for measuring force is newton, and unit for pressure is pascal. Certain parameters can be formulated indirectly through other units. Speed, for example, is defined as unit of distance passed per unit of time, and acceleration ââ¬â as rate of change of speed per unit of time. Consequently, force that influences a body, may be calculated through known mass of the body, its beginning speed and rate of change of its speed under the influence of the force studied. Unfortunately, measurements can never provide us with absolute values of parameter we are interested in. Due to imperfection of both human perception, instruments for measurements and because of influence of unstable environmental conditions upon the instrument, body that is subjected to measurements and the person performing measurements, some imprecision will always be present. Additionally, when performing measurements, observer sometimes has to use estimated data because more or less precious values can not be obtained under current conditions. Or maybe high precision values of a given parameter are not important for the current task. Either way, estimation of measured data sometimes takes place, as well as certain ever-present
